Research Focus of the Lab:
Cathepsins, a heterogeneous group of lysosomal proteases are ubiquitously present in the nature. They are involved in physiological functions such as degradation and turnover of intracellular proteins, neo-vascularization, antigen presentation, pro hormone activation, sperm maturation, embryo implantation, cell growth and tissue homeostasis etc. These functions are altered in many pathological conditions including cancer and other ECM related pathologies. These proteases are overexpressed in malignancy and silencing their expression lead to: impaired pro-hormone and pro-enzyme activation, immune defects, osteoporosis, epidermal hyperplasia, abnormal bone development and dilated cardiomyopathy. These observations underscore the importance of their controlled expression for various life processes. Therefore, the major focus of my laboratory is to understand the molecular mechanism(s) responsible regulation of these proteases particularly cathepsin L and B in several malignancies including gliomas, leukemia and gall bladder carcinoma with the objective of using it as a therapeutic target. Another major focus of our laboratory is to identify the biomarkers for early detection of gall bladder and oral cancer using high-throughput genomic and proteomic approach. We are also studying the molecular mechanism underlying the upregulation of RNA binding protein hnRNPD in the pathogenesis of oral cancer. PTEN mutations are preferentially found in human glioblastoma, with reported frequencies ranging from 15% to 40%. So there is an ongoing investigation in the lab to understand the clinical significance of PTEN regulated genes in human primary glioblastoma cells in relation to p53 status and understand their potential utility as prognostic markers and therapeutic targets.
